OBJECTIVE:To analyze the distribution of weight average molecular weight (r) of four Angelica polysaccharides isolated from Angelica sinensis (Oliv.)Diels:AP Ⅰ,AP Ⅱ,AP Ⅲ,AP Ⅳ and determine their characters of electric charge,in order to provide a practical analytical method for studying the constituent activity relationship of polysaccharides.METHODS:Two kinds of high performance liquid chromatography methods were employed,including high performance gel filtration chromatography for r determination and high performance anion exchange chromatography for electric charge analysis.Improved phenol sulfuric acid method combined with ELISA reader were used for the polysaccharide fractions determination,and Sever blue G dye combined method for protein fractions as well.RESULTS:Each of the four Angelica polysaccharides,AP Ⅰ,AP Ⅱ,AP Ⅲ and AP Ⅳ,was composed of 4~5 polysaccharide constituents with different r distribution and characters of electric charge;and there were small amount of different free or combined proteins in the four Angelica polysaccharides.CONCLUSION:The four Angelica polysaccharides,AP Ⅰ,AP Ⅱ,AP Ⅲ and AP Ⅳ,are different from each other in r and character of electric charge;this method is practical and reliable for the study of constituent activity relationship of polysaccharides.

目的 :分析当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ各组分的重均相对分子质量分布及其电荷特性 ;探讨多糖类生物大分子的组效关系研究方法。方法 :采用高效凝胶过滤色谱法、阴离子交换色谱法、二极管阵列检测器进行检测 ;改良苯酚 硫酸法测定糖分部 ;ServaBlue G染料结合法测定蛋白质分部。结果 :当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ分别由 4~ 5个具有不同重均相对分子质量分布及电荷特性的多糖组分组成 ;4种多糖中还分别含有一定量的游离或结合蛋白质。结论 :当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ为组成成分不同的多糖 ;该方法可为多糖组效关系研究提供简便、实用的分析方法。

AIM To isolate and to purify some polysaccharides fractions from Angelica sinensis (Oliv.) Diels and to analyze and to determine the characters of each fraction. METHODS Hot water extracting and ethanol precipitating method was employed to isolate total polysaccharide (named AP 0) from fresh roots of Angelica sinensis. By treating AP 0 with 80 g·kg -1 Cetyltrimethylammonium Bromid (CTAB), the AP 1 was obtained from the precipitate. AP 2 was obtained from the precipitate by treating the supernate left from preceding step with 1 g·kg -1 H 3BO 3, 2 mol·L NaOH. After treating the left supernate with acetatic acid and precipiting with ethanol, AP 3 was obtained. The amounts of total carbohydrates, proteins and uronic acids were determined with improved phenol sulfuric acid, Serva Blue G dye binding and m hydroxyldiphenyl methods, respectively. The monosaccharides contained in each polysaccharides were analysed by using paper chromatography and thin layer chromatography. High performance gel filtration chromatography and high performance anion exchange chromatography were employed to determine the relative molecular masses and charge characters of each polysaccharides. Infrared spectrophotometer was used to analyze the structures of glucoside bonds of each sample. RESULTS Four Angelica polysaccharides were obtained. The total carbohydrates, uronic acids and proteins of each Angelica polysaccharides were: 970 g·kg -1 , 210 g·kg -1 and 30 g·kg -1 of AP 0; 970 g·kg -1 , 172 g·kg -1 and 30 g·kg -1 of AP 1; 830 g·kg -1 , 257 g·kg -1 and 170 g·kg -1 of AP 2 ; 970 g·kg -1 , 86 g·kg -1 and 30 g·kg -1 of AP 3. The monosaccharides constituents of the Four Angelica polysaccharides were glucose, arabinose and uronic acids. 4 Angelica polysaccharides were composed of 5 fractions with different Mr and 4 fractions with different charge characters , respectively. Infrared spectrum suggested that the glucoside bonds be α D glucopyranose. CONCLUSION Methods of purification and analysis were effective and practical for the preparation of Angelica polysaccharides. 4 Angelica polysaccharides were different in their physico chemical characters.

Objective:To compare the release ability of water soluble polysaccharides in sporoderm broken and nonbroken spores of Ganoderma lucidum , and establish a comparatively correct method for the determination and analysis of polysaccharide contents in Chinese herbs.Method:The release ability of water soluble polysaccharides was determined on the basis of phenol sulfuric acid modification in different conditions.Result:The release ability of polysaccharides of sporoderm broken spores was much grerter than that of nonbroken spores; and the phenol sulfuric acid modified cation method proved excellent in accuracy and reproducibility, with a relative error less than 1.5%.Conclusion:The spores should be wall wracked if used as a nutriment, or for extraction and analysis of their effective components. The method can be successfully used for the determination of polysaccharide contents in Chinese herbs or nutriments. [
